Electrostatically Crosslinked Protein Ionogels for Sustained Drug Delivery

Abstract

Glioblastoma (GBM) is an aggressive brain tumor with limited treatment options. While PD-1 checkpoint inhibitors hold promise, systemic delivery is ineffective due to poor blood-brain barrier penetration and rapid clearance. Local delivery via hydrogels has been explored, but traditional hydrogels degrade quickly due to its weak mechanical strength and high water content, limiting sustained drug release. We propose a biodegradable, cross-linker-free protein ionogel that leverages electrostatic interactions between proteins and ionic liquids to enable controlled PD-1 release over months rather than weeks. Unlike conventional hydrogels, this tough, tissue-mimetic ionogel offers prolonged therapeutic action, reducing the need for frequent dosing. Beyond GBM, this platform could transform localized drug delivery, enabling long-term controlled release of biologics for cancer, regenerative medicine, and chronic disease management.

Receiving the PhRMA Foundation Faculty Starter Grant is invaluable in launching my new lab. It provides critical momentum to advance bold, high-risk ideas toward improving the delivery of complex biologics, such as proteins, nucleic acids, and cells.
